Background
The Toll-like receptor (TLR) family in mammals comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. The TLR family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. TLR3, 7, 8, and 9 are expressed in the intracellular organelles. TLR7 and TLR8 have a common ligand, viral single-stranded RNA. TLR3 is activated by viral double-stranded RNA. TLR9 responds to bacterial unmethylated CpG DNA. Ten human homologs of TLRs (TLR1-10) have been described. Following are the different antibodies included in the TLR Sampler Kit:
Intracellular flow analysis of TLR7 in human PBMCs using IMG-581A at 0.5 ug/ml. Shaded histogram is cells alone, green is rabbit IgG isotype control (Imgenex, Cat. No. 20304), red represents anti-TLR7 antibody. Imgenex's goat anti-rabbit PE (IMGENEX, 20303) was used for secondary.
Western blot analysis of TLR3 in lysates from untransfected 293 cells (lane A), 293 cells transfected with human TLR3 cDNA (lane B), and human intestine tissue lysate (lane C) using IMG-315A at 2 ug/ml.
Intracellular flow analysis of TLR9 in Ramos cells using 0.1 ug of IMG-305A. Shaded histogram represents Ramos cells without antibody; green represents isotype control (IMGENEX, 20109); red represents anti-TLR9 antibody. Imgenex's goat anti-mouse FITC (IMGENEX, 20102) was used for secondary.
